Activity 6.1Class 10 Science
Object:
Light is essential for photosynthesis
Materials Required:
A distracted potted plant with variegated leaves. (Variegated means leaves with
green and non-green areas, like leaves of Croton or money plant). Leaves can be
destarched by keeping the plant in a dark room for three days, Petri dish,
tripod stand, wire gauze, blotting paper, dropper, spirit lamp or burner, water
bath, 70% alcohol, iodine solution, translucent butter paper, soft pencil.
Procedure:
We will follow the below steps to perform this activity-
(i) First, we keep the plant in a dark room for three
days so that the starch gets used up.
(ii) Now Keep the destarched plant in sunlight for
about six hours.
(iii) Pluck a
leaf from this plant. It should be a variegated leaf.
(iv)
Place translucent butter paper over the
leaf.
(v)
Mark the green and non-green areas on it. We can make a separate diagram of the
leaf showing the pattern of variegation. Colourless areas indicate the absence
of chlorophyll.
(iv) Dip this leaf in boiling water for a few
minutes.
(v) Take the leaf out and now dip it in a beaker
containing alcohol.
(vi) Now place this beaker carefully in a water
bath and heat till the alcohol begins to boil (Alcohol is a highly inflammable
substance, take care) Alcohol decolourizes the leaf completely.
(vii) Take out the decolorized leaf dip in water to
remove the alcohol.
(viii) Spread the leaf in a Petri dish and put a few drops of iodine solution. After 2-3 minutes rinse off excess iodine
solution, and observe.

Observation:
This leaf after treatment with iodine shows two types of patches. Some areas
are bluish-black while others are yellowish. Bluish black areas show the presence
of starch. The yellowish area colourless areas devoid of starch. When these
patches are compared with the diagram of the leaf showing variegation pattern,
it becomes of the leaf showing variegation pattern, it becomes clear that bluish-green areas are the ones which were previously green. Non-green areas remain
colourless or yellowish.
Inference/conclusions:
Now it can be inferred that only the chlorophyll-containing areas produce
starch. It proves that chlorophyll is necessary for photosynthesis.
Precautions:
(i) Destarch the potted plant properly.
(ii)Make the pattern of variegation on butter
paper. It should be compared with the leaf after the iodine test.
(iii) Alcohol should be heated in a water bath only.
(iv)
We must take care when using alcohol.
